Biocolor Fastin elastin Assay
利用染料結合的方法來定量檢測釋放到組織培養(yǎng)液或從生物材料中提取的彈性蛋白。
應用范圍:樣本可來源于細胞培養(yǎng)基和含細胞外基質成分的組織提取物(如彈性軟骨、主動脈、皮膚等),可檢測可溶性彈性蛋白原和不可溶性彈性蛋白(抽提后以可溶性彈性蛋白多肽的形式檢測)。
樣本要求:樣本體積5-100ul,包含5-75ug的彈性蛋白
實驗操作步驟
彈性蛋白的分離:用彈性蛋白沉淀劑處理樣本,使彈性蛋白沉淀下來。
彈性蛋白與染料結合:裝有沉淀的彈性蛋白管中,加染料和反應液,反應60min以上
彈性蛋白-染料復合物獲得:去掉未結合的染料
復合物染料的釋放:添加染料釋放劑,使彈性蛋白-染料復合物中的染料釋放出來
釋放出的染料檢測:繪制標準曲線,513nm或405nm處定量檢測
